Overproduction of tryptophanyl-tRNA synthetase relieves transcription termination at the Escherichia coli tryptophan operon attenuator.
Overproduction of tryptophanyl-tRNA synthetase increased trp operon expression by reducing transcription termination at the trp attenuator. The total cellular level of charged tRNATrp was not affected by increased levels of the synthetase. We propose that excess synthetase binds charged tRNATrp and reduces the concentration available for translation.
متن کاملUse of pIVEX plasmids for protein overproduction in Escherichia coli
BACKGROUND The pIVEX plasmids are vectors optimized for expression in the Rapid Translation System (RTS) cell-free system under control of bacteriophage T7 transcription elements. Even if these plasmids are intended for use in vitro, it is usually worthwhile to compare both cell-free and bacterial expression from the same genetic construct. However, some RTS users encountered problems when they...
متن کاملMultiple feedback loops are key to a robust dynamic performance of tryptophan regulation in Escherichia coli.
Living systems must adapt quickly and stably to uncertain environments. A common theme in cellular regulation is the presence of multiple feedback loops in the network. An example of such a feedback structure is regulation of tryptophan concentration in Escherichia coli. Here, three distinct feedback mechanisms, namely genetic regulation, mRNA attenuation and enzyme inhibition, regulate tryptop...
متن کاملSuccinate Overproduction: A Case Study of Computational Strain Design Using a Comprehensive Escherichia coli Kinetic Model
Computational strain-design prediction accuracy has been the focus for many recent efforts through the selective integration of kinetic information into metabolic models. In general, kinetic model prediction quality is determined by the range and scope of genetic and/or environmental perturbations used during parameterization. In this effort, we apply the k-OptForce procedure on a kinetic model...
متن کاملMolecular Characterization and Antimicrobial Resistance of Uropathogenic Escherichia coli
Background:Urinary Tract Infections (UTIs) are the most common infectious diseases in childhood. The Uropathogenic Escherichia coli (UPEC) strains account for as much as 80% of UTIs. Objective:From a clinical perspective, it is important to know which virulence factors and antibiotic resistance properties are present in UPEC strains in pediatrics. Therefore, this study was carried out...
